The last time i ran our engine and took our boat out for a run was the 7th March.
What should be checked before starting the engine and taking the boat out again? Being shaft drive, I'm concerned about growth on the prop shaft, am I right to be?

Yes and no, yes there is likely to be some growth, but it’s not much to worry about. If it doesn’t come off when in motion, you can always clean it off either in or out of the water. Fouling on your prop will have more affect on performance, but again that can be cleaned.

Cutlas bearing etc will be fine.

just do normal checks. Oil. Coolant. No obvious leaks. Sea water filters. Nothing out of place.

Start it up and have a look a min or 2 later to check nothing strange is going on.

briefly excercise fwd / reverse / bowThruster / steering then go have fun

Then let it get hot and do your checks again.
