Right by the town. Lots of charter boats stern to. For the location, check on a map & it's the northern most of the harbours.
 
Very difficult to find a free space during the summer unless you pay the local agent (and pay them!).
Provided you find a space, it's near everything. The old town, the new town, restaurants, tavernas, cafes, shops, supermarkets etc, etc.
The main harbour is next to it.
Be careful entering mandraki. There are some rocks near the entrance. Check the pilot.
 
Nightmare place. The charter companies block spaces, unless you have arranged a space with an agent you are unlikely to get one, and if you do fouled anchors are common. If you do find an empty space you are likely to be forced out out of it by agents who have "reserved" it for somebody else, despite the fact that it is a public harbour (and not a marina). Rhodes was paid by the EU to build a proper marina with our tax money. They took the cash and never finished the marina. My advice is boycott Rhodes until there is a proper safe marina, or go only out of season, when you might find space, otherwise be prepared to anchor off. Some years ago I stayed in the unfinished "New Marina", but it was unsafe when a swell came in.
