Loctite 5331 is less viscous than Sikaflex so easily fills the threads on both sides. It can also be undone if you mess it up. As said earlier coating both male and female threads around the full circumference is foolproof, IMO.
Sikaflex is a brand. Are you referring to the specific Sika product mentioned on thread which is not the 291 seen in chandleries and used on deck fittings.

Shouldn't parallel thread fittings tighten down onto a sealing washer or something?
I'm about to tackle this with a TD seacock. I'm thinking of including an O ring at the base of the female threads as well as silicone sealant on the threads. To be sure, to be sure...

Any reason I shouldn't?

I can’t see it causing harm but it also won’t help, if it leaks it needs sorting not an oring which may well fail later. An oring is unlikely to seal on threads anyway. The TD instructions are pretty clear and work very well so I don’t see a reason to add things

There's a flat on the bottom of the female threads and on the ends of the skin fitting and hose tail. I was thinking an o ring in there, not on the threads.

There are several reports in this thread of leaking joints.

The only downside that I can see is the scope for positioning the seacock handle will be limited.

My point was that an oring is not the solution to those leaks, they need redoing and sorting properly.
