To repeat what I said previous in this thread, that seat you refer to is a wet locker (hence the drain in the bottom) why risk spoiling it's integrity by putting a hatch in there?
You also said (after I asked) that there was a glovebox type affiar in the bulkhead alongside this locker, now if as suspected this is a dry locker then why not create access through this?
There's no issue putting a 6" round inspection hatch where you've marked - I'd put one on each side to match. The only thing to consider are whether you have any fittings mounted behind the panel such as transom shower / water / drain tubes etc.
I honestly think you're worrying too much about minor issues - just get on and enjoy the boat!
