Holding tank fittings

I am currently installing a good quality holding tank, which I hope will not be smelly. But what is the best way to seal the various fittings that screw into it? The tank is plastic, and some of the screw in fittings are also plastic, whereas a couple are brass. Should I use plumbers tape to seal the threads? Or something else?

Also, the only place to fit the tank is about 1.5 metres above the Jabsco joker valve. Might it be advisable to fit a second non-return valve in the pipe downstream of the Jabsco to give the joker some help against the slightly higher head of liquid? Or is that just asking for extra blockages?
 
Due to a cock-up in the design of my holding tank, entirely my fault, the outlet finished up in the wrong position. I installed a 1.5 inch plastic skin fitting instead. This comes with a neoprene rubber washer, which I fitted against the flange inside the tank and sealed the whole thing with Sikaflex 291. That was more than 5 years ago and it has not leaked or shown signs of distress.

A head of 1.5 metres is not a lot, to the top of the loop in mine and most others it is around a metre. Additional head should assist the joker valve to close, as the rubber in the tri-cuspid part tends to open a little with age, which pushes the lips apart. A bit more head might help to keep them closed.
