Before dismantling lockers and crawling into the void, does it are much difference which way round the cooling water connections from an engine to a calorifier go?
The cylinder is horizontal I think.

Cunningly I made life a little more challenging by not marking the 2 pipes when the old engine came out.......

Convention is hotter water in higher up, cooler out lower down that way the natural thermo-syphon effect assists the water flow, plus the temperature difference outside the coil will be more event - the hot water inside the tank will rise to the top, where the hotter water is entering and vice versa.

If your connections are horizontal can't see it will make any difference at all.

Best in at the bottom and out at the top to avoid air locks, most units come new with in/out labels on the coil(s)

That is probably the case with an indirectly cooled engine that has a relatively powerful pump to drive the water around. With my raw water cooled installation I found that the thermo siphon effect pretty much cancelled the puny efforts of the little in-line electric pump I was using, so connecting your way around was not working. Using superheat6k's routing did cause some airlock problems that were solved by back-filling from the calorifier end but it worked well once full.
